So if a Tzimisce removed everything in his brain and nervous system regarding sleep and tiredness, he would become imune to quite a few disciplines  just saying

Not really possible. Even the simplest animals have activity cycles. Actually, even bacteria do. Not to mention some organisms on higher organisational tiers. Plants have photosynthetic cycles in metabolism (OK, forced by light/dark but still).
To cut a lengthy essay short, activity has to be mirrored by rest. An animal unable to rest would simply die. Same happens in humans with long enough sleep deprivation. Sleep is necessary to remove harmful metabolites from the brain, to put it simply.
Theoretically speaking though, such a hypothetical animal might or might not be susceptible to chemical sleep induction. Depends what made it sleepless. If lack of melanin receptor/other sleep related receptors, then yes. If something else... God only knows. Not possible to answer without thorough research.

Adding spikes/body armour adds function. But what moves those is Vitae, not dead nervous system. And Disciplines affect Vitae...

I don't think that would work all that well because Bloodlines has very different srengths than TES games. Bethesda is good at crafting worlds, but Fat Larry has more personality than all the NPCs in Skyrim combined.

I'd be interested to see Bethesda do a VtM game, but I wouldn't want it to be Bloodlines 2.

I'd love to see it happen in Chicago or NYC.
